About Hardware connections :

For ISP connection you need to make jumper CMD, CLK, DAT0, VCC, & VCCQ. Sometimes you need to make jumper only CMD, CLK, DAT0 & USB to device, But Some devices need high voltage through which don’t come through USB. So for that devices need to make jumper all connections CMD, CLK, DAT0, VCC, & VCCQ.
